Academy Project: Investigating feasibility of scaling up crystallization of a membrane protein

Objective
The primary objective of this project is to investigate the feasibility of scaling up a protein stabilization technique for the crystallization of membrane proteins in 2D sheets. This ambitious endeavor requires a significant increase in volume, alteration of chemicals, and innovative utilization of equipment. Through a comprehensive experimental and theoretical study, the aim is to assess the viability of scaling up this crucial process, which holds immense potential for advancing biomimetic membrane development.

Tasks
Literature study on membrane proteins, 2D sheet formation, and detergent study.

Experimental tasks:

  • Volume Scalability Assessment: Determine the optimal scale-up factors to maintain protein integrity and sheet morphology.
  • Chemical Modification and Compatibility Testing: Investigate the impact of altering chemicals and concentrations on the stability and ability to form crystals of membrane proteins. Identify suitable chemical formulations that facilitate scalability without compromising the structural integrity of the 2D sheets.
  • Equipment Innovation and Optimization: Explore innovative approaches to adapt existing laboratory equipment and develop a setup capable of accommodating larger volumes and facilitating the formation of uniform 2D protein sheets. Optimize equipment performance to ensure reproducibility and scalability across different experimental conditions.
  • Characterization: Employ advanced analytical techniques, such as transmission electron microscopy (TEM) to characterize the morphology and structure of the scaled-up 2D protein sheets.

Student Opportunities and Profile
