Master thesis project: Developing new terminal sterilization technologies for liquid pharmaceuticals
Project description
In the production of finished dosage pharmaceuticals, preventing microbial contamination is crucial to maintaining the safety of the patients. The gold standard for preventing this kind of contamination is terminal sterilization by heat. However, there are currently no liquid protein-based pharmaceuticals that are terminally sterilized on the market. This is due to the high amounts of stress introduced by sterilization processes to the product leading to a significant loss of quality, function, or both. This project aims to investigate the possibility of applying terminal sterilization to a finished dosage pharmaceutical through developing a less stressing sterilization process. The core idea for this process is to use centrifugation to concentrate endospores in a small portion of the container, before delivering a focused sterilization dose to the microbes using a high-powered visible light or NIR laser.
Key Challenges
- Developing a new technology to terminally sterilize protein-based liquid pharmaceuticals.
- Ensuring a comprehensive and validated concentration and subsequently inactivation of endospores.
- Characterizing the effects of high-powered NIR and/or visual light lasers on the active pharmaceutical ingredient.
Student profile
We are looking for students who are proficient in one or more of the following subjects:
- Microbiological work
- Protein characterization
- Laser optics
- Spectroscopy
- Data analysis
Within these fields the student will be given an opportunity to investigate some of the following topics:
- The effects of high-powered lasers on API (active pharmaceutical ingredient)
- The killing efficacy of high-powered lasers against endospores
- The characterization of centrifugal concentration of endospores & endospore analogs through fluorescence microscopy (and potentially computational fluid dynamics)
The specifics of the project can be discussed based on the student’s competencies and interests.
